In a depletion workflow, first a depleting antibody in vivo is administered and then the remaining cell population is analyzed by flow cytometry. Problems arise when the validation antibody recognizes the same or an overlapping epitope as the depletion antibody. This can lead to:

- false-negative detection of residual cells
- incorrect interpretation of depletion efficiency
