Navinci’s NaveniFlex™ Kits are in situ proximity ligation assay (PLA) kits designed for the precise detection of proteins, protein-protein interactions, and post-translational modifications within cells and tissue samples. These kits utilize proprietary Navenibodies as secondary detection reagents, which, when in close proximity, initiate a rolling circle amplification, resulting in a quantifiable fluorescent or chromogenic signal. This technology enables researchers to visualize and quantify molecular interactions with high specificity and sensitivity, even for low-abundance targets, while preserving the spatial context of the sample.

NaveniFlex™ Cell
The NaveniFlex™ Cell PLA is an in situ proximity ligation assay kit optimized for cell samples (FFPE and fresh formaldehyde-fixated cells). It enables in situ fluorescence visualization of proteins, protein – protein interactions, or protein modifications in cell samples. It allows to analyze even the most low-abundance protein targets with precision and ensures high specificity. The obtained data is quantifiable. High throughput screening with 96 well plate protocol is possible.

Naveni™TriFlex Cell is designed for fluorescence detection of two proteins in a free and complex state. Based on the Naveni™ Proximity Ligation Technology, the in situ kit provides accurate data with precise staining of total protein A, total protein B, and the interaction AB between the two proteins.

Naveni™ TriFlex Cell - Workflow and Sample Types
Naveni™ TriFlex relies on two user-determined primary antibodies against the targets of interest, and on proprietary TriFlex Navenibodies (in orange), which are antibody-based proximity reagents. Only proteins located at <40 nm distance are recorded as interacting. Naveni™ TriFlex detects total protein A (i.e., both free and in complex with B, yellow), total protein B (maroon), and the AB interaction (blue). The detected A, B and AB signals are amplified and generate fluorescent readout in three channels corresponding to each protein pool.

2 - Co-localization is not a synonym to protein interaction
NaveniTMTriFlex Cell was used to simultaneously detect total COX1 and GM130, and their potential interaction. This assay was used as a biological negative control, as COX1 and GM130 belong to different cell compartments and thus do not interact, what is visualized using the Cy5 channel. Note that in the merged image, however, COX1 and GM130 appear partially co-localizing (white) where signals are particularly dense.

PLA Kits optimized for Tissues
NaveniFlex™ Tissue
NaveniFlex™ Tissue kits are specifically optimized for tissue samples (FFPE, frozen) to improve sensitivity and to overcome unspecific bindings and common background challenges. The kits deliver highly reproducible and accurate results in tissue samples, even for the least abundant protein interactions. Compatible with traditional immunostaining equipment (fluorescent readout).
EASY PROCEDURE: The workflow is basically identical to a traditional immunostaining!

NaveniFlex™ Tissue solves common tissue staining problems
PROBLEM:
A commercial in situ proximity ligation kit X (a, b) was used for Podocalyxin/Ezrin staining in human FFPE breast cancer tissue. The kit produces similar stain in both the positive (a) and the technical negative control (b) due to high background (hazy red staining) which obscures proximity signal.
SOLUTION:
NaveniFlex™ Tissue visualizes the Podocalyxin/Ezrin interactions clearly (discrete red specks), free of background (c), and leaves the technical negative control blank (d) in FFPE breast cancer tissue. A picture does say more than a thousand words!
The NaveniFlex™ Tissue kit includes two Navenibodies conjugated to proprietary oligo arms (orange antibodies). Only if the Navenibodies are in close proximity, they generate a rolling circle amplification reaction, leading to a strong and distinct dot that is easily quantifiable.
