OZ Biosciences PolyMag Premium - Upgrade Magnetofection for Primary & Hard-to-Transfect Cells

PolyMag Premium™ is the new generation magnetic nanoparticle transfection reagent from OZ Biosciences, engineered as a powerful upgrade of the classic PolyMag. Specifically developed for demanding cell systems, it delivers high gene transfer efficiency in primary cells and hard-to-transfect models while preserving viability. Using Magnetofection™ technology, nucleic acid–loaded magnetic particles are rapidly concentrated onto the cell surface by a magnetic plate, greatly increasing contact time compared with diffusion-based lipid reagents.

Combined with an optimized Boost additive, PolyMag Premium achieves stronger expression, higher percentages of transfected cells and robust performance in serum-containing media with minimal toxicity and reduced DNA/RNA usage.

PolyMag Premium is the upgrade of the Classic PolyMag

PolyMag Premium is the upgraded version of the original PolyMag reagent. Side-by-side data (Vero, C6 and HEK293 cells) show that:

  • Mean fluorescence intensity is higher at equivalent DNA doses

PolyMag Premium outperforms Competitors (Fugene® HD, Lipofectamine® 3000)

PolyMag Premium outperforms both the previous PolyMag reagent and leading lipid-based competitors (Fugene® HD, Lipofectamine® 3000) in terms of percentage of transfected cells and in terms of fluorescence intensity.

Hard-to-transfect cell lines

In C2C12, RAW264.7 and DC2.4 cells, GFP-encoding DNA was complexed either with PolyMag Premium (with or without Boost) or with competing reagents (FG=Fugene® HD, LF=Lipofectamine® 3000) according to their respective protocols, and transfection efficiency was assessed after 48 hours.

Across all three cell lines, PolyMag Premium alone achieved a higher percentage of GFP-positive cells than the lipid-based reagents, and also yielded greater fluorescence intensity per cell, reflecting stronger expression. When the Boost additive was included, PolyMag Premium delivered an additional increase in signal and/or transfection rate, depending on the specific cell type.

Primary mesenchymal stem cells (MSC)

In mesenchymal stem cells (MSCs), a highly relevant yet difficult primary model, PolyMag Premium with or without Boost clearly outperformed Lipofectamine 3000, achieving higher transfection efficiency as measured by the percentage of GFP-positive cells and markedly higher mean fluorescence—especially when Boost was added—indicating robust gene expression in each transfected cell.

Gene Silencing with PolyMag Premium

PolyMag Premium allows silencing gene expression mediated by siRNA.

PolyMag Gene Silencing

Efficiency of gene silencing with PolyMag Premium: NIH-3T3 (A) and C2C12 cells (B) expressing GFP were transfected with PolyMag Premium and ranging doses of siRNA-GFP. Gene silencing using siRNA-GFP was monitored 72H after under fluorescence microscopy.
